Proteomics


Proteomics is the systematic, large-scale analysis of proteins from a cell, tissue, biological fluid, or organism under a specific, defined set of conditions. It is able to characterise variations to a protein profile in response to environmental stimuli, differences in protein states between diseased and control systems, the spatial distribution of proteins and changes over time.

The discipline utilises three basic technological areas that have seen dramatic development over the past couple of decades. These include a method to fractionate complex protein or peptide mixtures, high-resolution mass spectrometry to acquire the data, and bioinformatic systems to analyse the large amount of generated spectral data.

Proteomics research is widely applied in many scientific fields such as disease research (i.e., cancer and diabetes), diagnostic biomarker discovery, plant and animal phenotyping, drug discovery, microbiology and antibody profiling.

A typical proteomics study involves the following steps:

(B) Protein extraction is critical to obtain a representative sampling of proteins under the defined conditions. (C) Sample complexity is reduced by gel or liquid chromatography separation of proteins prior to enzymatic digestion and MS analysis (D). (E) The observed MS spectra from each sample are bioinformatically matched to protein libraries to obtain protein identifications. (F) Finally, proteins of interest are validated by independent techniques, usually involving antibody-based methods such as western blotting or ELISA.
